Pancreatic cancer is the fifth leading cause of cancer death in the US. High mortality associated with pancreatic cancer arises partly due to inability to detect this aggressive cancer early. None of the biomarkers of pancreatic cancer that have been identified to date is sufficiently predictive to be useful as a diagnostic or screening test. We hypothesized that a panel comprised of multiple cancer-associated biomarkers may facilitate earlier detection of pancreatic cancer. In this study we utilized a novel protein array system (xMAP® for Multianalyte Profiling; Luminex Corp., Austin, TX), which allows for simultaneous quantitation of up to 100 soluble analytes in one sample. We evaluated the diagnostic efficacy of a multimarker panel using 44 serological markers, including cytokines, chemokines, adhesion molecules, and hormones. Sera from 100 patients with pancreatic cancer and 400 healthy controls were screened. Our data have demonstrated that patients with pancreatic cancer have significantly different expression patterns of several serum biomarkers compare to healthy controls. Our statistical algorithm demonstrated the best performance as compared to other methods resulting in 97% correctly classified cancers with a sensitivity of 95% at a specificity of 98 %. The selected multimarker panel offering highest diagnostic power was comprised of 10 biomarkers, including CA19-9, CA-125, and others. To our knowledge, this study offered the largest panel of serological markers to be examined simultaneously in pancreatic cancer. Our results have demonstrated that analysis of multiple serum cytokines using a novel LabMAPTM technology is a promising new approach for development of a diagnostic assay for pancreatic cancer.
